چکیده

Background: Constitutive activity is central to G protein-coupled receptor signaling still the mechanisms underlying it are unknown. Results: the ghrelin receptor monomer reconstituted in a lipid disc activates Gq without agonist and recruits arrestin in a liganddependent manner. Conclusion: high constitutive activity is an intrinsic property of the ghrelin receptor. Significance: this is the first demonstration that the ghrelin receptor has all the determinants for constitutive activity and ligand-regulated internalization.

A monomeric G protein-coupled receptor isolated in a high-density lipoprotein particle efficiently activates its G protein.

G protein-coupled receptors (GPCRs) respond to a diverse array of ligands, mediating cellular responses to hormones and neurotransmitters, as well as the senses of smell and taste. The structures of the GPCR rhodopsin and several G proteins have been determined by x-ray crystallography, yet the organization of the signaling complex between GPCRs and G proteins is poorly understood. The observat...

Purification and Functional Reconstitution of Monomeric -Opioid Receptors

Despite extensive characterization of the -opioid receptor (MOR), the biochemical properties of the isolated receptor remain unclear. In light of recent reports, we proposed that the monomeric form ofMOR can activate G proteins and be subject to allosteric regulation. A -opioid receptor fused to yellow fluorescent protein (YMOR) was constructed and expressed in insect cells. YMOR binds ligands ...

The Functional Significance of G Protein-coupled Receptor Dimerization

THE FUNCTIONAL SIGNIFICANCE OF G PROTEIN-COUPLED RECEPTOR DIMERIZATION by Matthew R. Whorton Chair: Roger K. Sunahara G protein-coupled receptors (GPCRs) are seven transmembrane domain proteins that transduce a diverse array extracellular signals across the plasma membrane and couple to the heterotrimeric family of G proteins. Like most intrinsic membrane proteins, GPCRs are capable of oligomer...

Recombinant Human Melatonin Receptor MT1 Isolated in Mixed Detergents Shows Pharmacology Similar to That in Mammalian Cell Membranes

The human melatonin MT1 receptor-belonging to the large family of G protein-coupled receptors (GPCRs)-plays a key role in circadian rhythm regulation and is notably involved in sleep disorders and depression. Structural and functional information at the molecular level are highly desired for fine characterization of this receptor; however, adequate techniques for isolating soluble MT1 material ...

Expression, Functional Characterization, and Solid-State NMR Investigation of the G Protein-Coupled GHS Receptor in Bilayer Membranes

The expression, functional reconstitution and first NMR characterization of the human growth hormone secretagogue (GHS) receptor reconstituted into either DMPC or POPC membranes is described. The receptor was expressed in E. coli. refolded, and reconstituted into bilayer membranes. The molecule was characterized by 15N and 13C solid-state NMR spectroscopy in the absence and in the presence of i...
